Myeloperoxidase (MPO) is an enzyme of paramount importance within the canine immune system, specifically within neutrophils, the body's first line of defense against invading pathogens. MPO is housed primarily in the azurophilic granules of these specialized white blood cells. It serves as a critical component of the canine innate immune response.
Structurally, MPO is a heme-containing enzyme, which means it contains iron atoms at its core. When neutrophils encounter microbial invaders, they unleash MPO into the surrounding environment as part of their antimicrobial weaponry. MPO's primary function is to catalyze the production of hypochlorous acid (HOCl) from hydrogen peroxide (H2O2) and chloride ions (Cl-). HOCl is a potent and reactive antimicrobial agent capable of swiftly neutralizing a broad spectrum of microorganisms, including bacteria, fungi, and viruses.
MPO's mechanism of action involves the generation of reactive oxygen species (ROS), such as superoxide radicals and hydrogen peroxide, which are toxic to pathogens. Additionally, MPO promotes the conversion of chloride ions into hypochlorous acid, which exhibits potent antimicrobial properties, further enhancing the neutrophils' ability to eliminate invaders.
