Bintrafusp alfa (M7824) is a bifunctional fusion protein composed of the extracellular domain of the human TGF-β receptor II (TGF-βRII or TGF-β ‘trap’) fused via a flexible linker to the C-terminus of each heavy chain of an IgG1 antibody blocking PD-L1 (anti-PD-L1). Bintrafusp alfa is more effectively suppressed tumor growth and metastasis than either a TGF-β trap or an anti-PD-L1 antibody alone. Bintrafusp alfa also inhibited spontaneous metastasis in multiple preclinical models, presumably due to its suppressive effect on epithelial-to-mesenchymal transition
