Cell Sorting & Processing

The results of TCE functional assays depend not only on the candidate molecule itself but also significantly on the state of the effector and target cells. T-cell composition, CD4/CD8 ratio, cellular activation state, target-antigen expression, and the effector-to-target (E:T) ratio can all alter the final cytotoxicity and cytokine response. Therefore, before comparing multiple TCE candidates, a stable and highly reproducible cell system must first be established.

T-Cell Preparation and Enrichment

Enrichable T-cell subsets: CD3⁺ T cells; CD4⁺ T cells; CD8⁺ T cells.

For T-cell isolation, a negative-selection strategy is recommended. Positive selection uses anti-CD3 or anti-CD4/CD8 antibodies to directly label T cells; antibody binding may cross-link the TCR complex, causing pre-activation of T cells and producing high background signals that interfere with the assessment of true candidate activity. Negative selection removes monocytes, B cells, NK cells, and other non-target cells, leaving T cells in a resting state that more closely approximates the in vivo situation. The reagent should feature mild binding characteristics — uniformly sized, nanoscale magnetic beads paired with a specific antibody cocktail can complete isolation within 30–45 minutes, yielding resting T cells with >95% purity and >90% viability.

After isolation, purity and viability must be confirmed by flow cytometry, and resting-state markers checked (CD69 baseline should be negative). If both CD4⁺ and CD8⁺ subsets are required in the same experiment, positive-selection beads may be used to label them separately, but the cells should rest for 30–60 minutes before functional assays.

Compatibility of positive selection with flow restaining: Epitopes occupied by positive-selection antibodies compete with subsequent CD4/CD8 restaining antibodies, weakening or even ablating subset-gating signals in activation and exhaustion readouts. The protocol must explicitly note that the selection-bead clones and restaining antibodies must use non-competing clones or separated fluorescence channels. "Compatibility of selection reagents with multicolor flow panels" should be delivered as a formal validated item of the reagent product.

Target-Cell Preparation

Target cells likewise require phenotypic and antigen-expression confirmation, so that antigen-positive, antigen-negative, and varying antigen-density conditions can be properly defined. Use low-passage (<20) tumor cell lines; record target-antigen density by ABC quantitation before each experiment to avoid false-negative results caused by antigen loss. If different candidates are tested with T cells from different states, or if target-cell antigen expression differs markedly between batches, the observed "candidate differences" may partly originate from the experimental system itself rather than from the TCE molecules.

In practice, cryopreserved PBMCs should be rested overnight after thawing before sorting, and treated with DNase I to prevent cell clumping.

Blinatumomab Reference Data

Effector cells: CD3⁺ T cells obtained by negative selection from healthy-donor PBMCs, with >95% purity, >90% viability, and negative CD69 baseline. At least 3 donors are recommended. [1][2]

Target cells: CD19⁺ Raji or Nalm6 cells (B-cell lymphoma), low passage (<20); confirm high CD19 expression and stable ABC values before each experiment. Prepare K562 (CD19⁻) as the antigen-negative control. [1]

Effector-to-target ratio (E:T): 10:1 recommended; for titration, test 1:1, 5:1, 10:1, and 20:1. [1][2]

Note: Blinatumomab is Fc-free and does not mediate nonspecific activation through FcγR; however, residual monocytes in PBMCs may generate background cytokines during long-term co-culture, so PBMC experiments should include controls without target cells or with a non-targeting TCE.

Data Validation

GeneMedi's SOLIDEX®-ISOEx negative-selection kits are fully compatible with the epcoritamab functional assay system, yielding resting T cells with >95% purity and >95% viability and minimal background activation, providing high-purity, high-viability cells for TCE functional evaluation. In terms of sorting quality and viability, GeneMedi negative-selection kits perform on par with competitors. The real difference appears in the resting state and surface-marker integrity: the GeneMedi antibody cocktail is specifically optimized to cover non-target cells more comprehensively, leaving fewer residual non-T cells and therefore producing lower CD69/CD25 baselines on post-sort T cells.
