SOLIDEX®-ISOEx L Column (Cat No.:GMP-ISOEx-Column-L)

GeneMedi's SOLIDEX®-ISOEx L Column is developed for the separation of cells labeled with superparamagnetic nanobeads, enabling rapid and gentle separation of flowing cells while ensuring stable cell status and maintenance of activity.

This product is compatible with all SOLIDEX®-ISOEx Nanobeads in GeneMedi's column-based (ISOEx-Enhanced) system. Due to the small size of SOLIDEX®-ISOEx Nanobeads (Column-Based), a high-intensity magnetic field is required to retain the labeled cells. The column needs to be placed in a magnetic rack, where a strong magnetic field separates the target cells. The SOLIDEX®-ISOEx L Column is compatible with all magnetic separation stands and racks on the market.

When used in combination with SOLIDEX®-ISOEx Nanobeads, it can yield target cells with high purity, high recovery rate, and minimal labeling. The separated cells maintain good biological functions and can directly proceed to downstream experimental processes, such as flow cytometry analysis, cell culture, or functional assays. Furthermore, this product ensures experimental consistency and performance stability while, furthermore, this product ensures performance stability while providing excellent cost-effectiveness, helping users control experimental costs.

When used in conjunction with a SOLIDEX®-ISOEx Magnetic Separator (such as the MX1 or MX8), the optimized matrix within the column generates a high-gradient magnetic field powerful enough to efficiently capture cells even with minimal SOLIDEX®-ISOEx nanobead labeling. It is suitable for a diverse array of applications, ranging from small-scale research to the positive selection of rare cells and the depletion of strongly labeled cells.

A. Before adding the next buffer, ensure the previous buffer has completely flowed through (indicated by the absence of continuous dripping from the bottom of the separation column), but do not allow the column to run dry.

Comparative evaluation of the overall performance of GeneMedi's SOLIDEX®-ISOEx L Column and Company M's Column in the isolation of TCR alpha-beta (abT)+ cells. The Company M's Column yielded slightly higher numbers of bound cells and flow-through cells. Given the minor magnitude of this difference and the absence of coordinated advantages across multiple key performance parameters, it may arise from inherent variability in the isolation procedure.

The positive cell populations obtained from both columns exhibited comparable purity and cell viability, both meeting expected standards. In summary, the GeneMedi SOLIDEX®-ISOEx L Column demonstrates equivalent cell isolation efficacy to the Company M's Column. The SOLIDEX®-ISOEx L Column provides a reliable and effective isolation solution.

As shown, both columns achieved high purity of target cells. The Company M's Column (A) yielded slightly higher absolute numbers of bound cells and flow-through cells, but showed no significant differences in cell purity or viability compared to the GeneMedi 's SOLIDEX®-ISOEx L Column (B).

Effective Performance of GeneMedi's SOLIDEX®-ISOEx L Column for Cell isolation

GeneMedi's SOLIDEX®-ISOEx L Column exhibits excellent cell isolation performance and stable batch consistency. Taking the sorting of CD3+ T cells from cryopreserved PBMCs as an example, the A and B batches (n=2) of this column achieved post-sorting purity, viability, and recovery rates for CD3+ T cells all exceeding 90%. Experimental results demonstrate that GeneMedi's SOLIDEX®-ISOEx L Column performs comparably to the isolation column from Company M, providing a reliable and efficient solution for cell isolation.

The primary difference lies in their maximum cell loading capacity.

Q3: Can the SOLIDEX®-ISOEx columns be washed and reused?
No, they are intended for single use only.

Reusing columns poses a high risk of cross-contamination. Additionally, repeated washing alters the column's internal matrix, significantly compromising purity, recovery rate, and flow rate—leading to unreliable results.


Q4: Are the columns sterile? Do I need to sterilize them before use?
Yes, the columns are strictly sterilized and individually packaged. They are ready to use directly in a laminar flow hood or biosafety cabinet without additional sterilization.

However, always equilibrate (rinse) the column with the recommended buffer according to the protocol before applying your cell suspension.


Q5: What should I do if the column flow rate slows down or gets clogged?
Clogging is usually caused by cell clumps or exceeding capacity. We recommend these preventative measures:
  1. Filtration: Always pass your cell suspension through a 30 µm or 70 µm cell strainer before loading.
  2. Nuclease Treatment: If viscosity is high due to free DNA from dead cells, treat the suspension with DNase I.
  3. Capacity Check: Ensure neither the total cell number nor the labeled target cell number exceeds the column's specified limits.


Q6: Are these columns compatible with standard magnetic separators currently on the market?
