    Pathogen Information

Canine adenovirus type II (CAdV-2) is a member of the Adenoviridae family, a group of non-enveloped, double-stranded DNA viruses that can infect a wide range of hosts including humans, birds, and mammals. The virus primarily infects dogs, but it has also been reported in other canids such as wolves, foxes, and coyotes.

CAdV-2 is typically transmitted through contact with infected bodily fluids or tissues, such as urine, saliva, nasal secretions, and feces. The virus can also be spread by contaminated food or water. Once inside the host, the virus replicates in the respiratory tract, tonsils, liver, and spleen, with peak viral shedding occurring in the first few days after infection.

Infectious canine hepatitis (ICH) is the most common disease associated with CAdV-2 infection in dogs. ICH is a highly contagious and often fatal disease that is characterized by fever, vomiting, diarrhea, abdominal pain, and jaundice. The virus targets hepatocytes, causing inflammation and necrosis in the liver, which can lead to liver failure and death. The onset of clinical signs of ICH typically occurs within 4-7 days post-infection and can last for up to 10 days.

CAdV-2 can also cause respiratory infections, conjunctivitis, and kennel cough in dogs. Respiratory infections caused by CAdV-2 often present with clinical signs such as coughing, sneezing, nasal discharge, and pneumonia. Conjunctivitis, or inflammation of the conjunctiva, can be a primary manifestation of CAdV-2 infection or occur as a secondary complication of other diseases such as ICH. Kennel cough, a common respiratory disease in dogs, is often caused by a combination of CAdV-2 and other respiratory pathogens such as Bordetella bronchiseptica.

Diagnostic methods for CAdV-2 include virus isolation, serology, ELISA, and PCR. Virus isolation, the gold standard for detecting CAdV-2, involves the inoculation of samples onto cultured cells followed by observation for cytopathic effect. Serological tests such as ELISA detect antibodies produced by the host in response to CAdV-2 infection. ELISA may also target the fiber protein or other viral proteins to detect the presence of the virus. PCR is a highly sensitive and specific technique that targets genes such as the early region 1A (E1A) or hexon for detection.

The virion structure of CAdV-2 consists of a capsid containing three major structural proteins: hexon, fiber, and penton base. The hexon protein forms the majority of the capsid and plays an important role in stabilizing the virion. The fiber protein is responsible for attaching the virion to host cell receptors, facilitating entry and infection. The penton base protein is involved in the assembly of the capsid and can activate host cell signaling pathways.

The early gene region of CAdV-2 is composed of the E1A and E1B genes, which are involved in viral replication and pathogenesis. The E1A gene is particularly important for the initiation of viral DNA synthesis and the activation of host cell cycle progression. E1B, on the other hand, inhibits host cell apoptosis and prolongs the lifespan of infected cells to enhance viral replication.

In conclusion, CAdV-2 is an important viral pathogen of dogs, causing a range of diseases from ICH to respiratory infections and kennel cough. The virus has a complex virion structure and genetic makeup that enable it to replicate efficiently and evade host immune responses. Diagnosis of CAdV-2 often involves a combination of virus isolation, serological tests, ELISA, and PCR. With improved understanding of the pathogenesis and molecular biology of CAdV-2, more effective prevention and treatment strategies can be developed to mitigate the impact of this important viral pathogen on animal health.
