Pathogen Information
THEV belongs to the Adenoviridae family, a group of non-enveloped double-stranded DNA viruses common in humans and other animals. THEV is classified as Avian Adenovirus Group II, which includes several adenovirus serotypes such as 4, 10, 11, and the recently identified serotype 13 (THEV).
THEV has been identified as the causative agent of a widespread infectious disease with high morbidity and mortality rates among turkeys. The disease was first observed in eastern Turkey during the 1970s and has emerged as a significant economic threat to the poultry industry worldwide. The clinical manifestations of THEV infection vary depending on the age and immune status of the host, the virulence of the virus, and environmental factors.
Young turkeys are particularly susceptible to infection and usually develop acute purulent enteritis, characterized by depression, diarrhea, hemorrhagic lesions, and dehydration. Mortality rates among affected birds range from 5-50%, depending on the severity of the disease and the effectiveness of supportive care. In contrast, older turkeys and recovered birds may develop a subclinical or mild form of the disease, with mild to no visible symptoms.
THEV's genome contains approximately 35 kilobase pairs and encodes for several structural and non-structural proteins. The major structural proteins include fiber, hexon, penton base, and protease, which interact to assemble the capsid and mediate viral entry and replication. The fiber protein in particular plays a crucial role in viral binding to host cell receptors and determines the tissue tropism of different adenovirus serotypes.
The diagnosis of THEV infection relies on various techniques, including virus isolation, electron microscopy, serology, and molecular methods such as PCR and qPCR. Virus isolation involves the inoculation of infected tissues onto cell cultures, followed by the detection of cytopathic effect. Transmission electron microscopy enables the visualization of viral particles in infected tissues, whereas serology employs the detection of antibodies against THEV.
Molecular methods such as PCR and qPCR have demonstrated higher sensitivity and specificity for detecting THEV, especially during the early stages of infection. These techniques target specific regions of the THEV genome, such as the hexon gene or fiber protein, and can detect low viral loads with high accuracy. Additionally, ELISA can be used to quantify the presence of antigens or antibodies in infected samples.
In conclusion, Turkey Hemorrhagic Enteritis Virus (THEV) is a significant pathogen that poses a substantial threat to the poultry industry worldwide. The disease caused by THEV is characterized by acute enteritis and hemorrhagic lesions, especially in young turkeys. THEV's genome encodes for several key structural and non-structural proteins, including fiber, hexon, penton base, and protease, which are instrumental in the virus's entry and replication. Diagnostic methods for detecting THEV involve various techniques, including virus isolation, electron microscopy, serology, and molecular methods such as PCR and qPCR.
