Protocol

  1. Preparation: Resuspend PBMCs in separation buffer.
  2. Labeling: Add the Isolation Cocktail and incubate for 15 minutes at 2-8°C(3).
  3. Washing: Wash cells with buffer to remove unbound antibodies(4).
  4. Bead Incubation: Add Anti-Biotin Nanobeads and incubate for 15 minutes at 2-8°C(5).

Efficient Isolation of NK Cells Using SOLIDEX™-ISOEx Untouched Human NK Cell Isolation Kit (Column-Based)

To determine the purity of isolated NK cells, CD56+ NK cells were magnetically sorted from human peripheral blood mononuclear cells (PBMCs). Cells before and after separation were stained with CD3-APC and CD56-PE antibodies and subsequently analyzed by flow cytometry. Results demonstrated a remarkable increase in CD56+ NK cell purity from 8.19% pre-isolated to 94.3% post-isolated, confirming highly effective NK cell enrichment.

FAQ

What are the sample requirements? Can I use frozen PBMCs?
The kit is optimized for human PBMCs. Both fresh and cryopreserved PBMCs can be used. However, for frozen samples, ensure high viability (>90%) and remove clumps/debris with a strainer before labeling to prevent column clogging.
