SOLIDEX™-ISOEx Indirect Human TCR alpha-beta (abT)+ Cell Depletion Kit (Column-based) (Cat No.:GM-Tg-hg-MP2610-dep-kit)
Achieve superior enrichment of untouched TCR γδT cells, NK cells, or B cells by efficiently removing Human TCR alpha-beta (abT)+ Cells from PBMC. This Depletion kit utilizes a high-performance Column-based strategy combined with indirect Magnetic Beads labeling. By effectively trapping the unwanted αβ T cells in the column, the flow-through fraction contains the target cells of interest. The result is High Purity non-TCR αβ cells with no antibody/bead residue and Zero Activation, perfectly preserving their physiological state.
SOLIDEX™-ISOEx Indirect Human TCR alpha-beta (abT)+ Cell Depletion Kit (Column-based) is a Indirect TCR alpha-beta (abT)+ Cell Depletion Kit (Column-based, Non-releasable)

Application
- TCR γδT Cell Research: Enrichment of untouched gamma-delta T cells for cancer immunotherapy studies.
- GvHD Research: Depletion of TCR αβ+ cells to study Graft-versus-Host Disease reduction in allogeneic transplantation models.
- NK Cell Analysis: Obtaining pure NK cell populations free from T cell interference for cytotoxicity assays.
- Chimerism Analysis: Isolating specific subsets for post-transplant monitoring.
Protocol
- Preparation: Prepare a single-cell suspension from Human PBMCs.
- Labeling: Incubate cells with the primary antibody cocktail targeting TCR αβ.
- Bead Binding: Add the Anti-Biotin/Secondary Nanobeads to bind the primary antibodies.
- Separation: Apply the suspension to a column placed in a magnetic field.
- Collection (Depletion): The unwanted TCR αβ+ cells are retained in the column. Collect the flow-through, which contains the untouched target cells (e.g., γδT cells, NK cells).

Efficient depletion of TCR alpha-beta (αβ)+ T cells from human PBMC using the SOLIDEX™-ISOEx Indirect Human TCR alpha-beta (αβ)+ Cell Depletion Kit (Column-based)
Catalog No. GM-Tg-hg-MP2610-dep-kit
This experiment was designed to evaluate the depletion efficiency of the SOLIDEX™-ISOEx Indirect Human TCR alpha-beta (αβ)+ Cell Depletion Kit (Column-based) for removing TCR alpha-beta (αβ)+ cells. Data analysis demonstrated the kit's excellent depletion performance. In the initial PBMC sample, TCR alpha-beta (αβ)+ cells accounted for 54.7%. After processing through the separation column, the residual TCR alpha-beta (αβ)+ cells in the flow-through fraction dramatically decreased to 0.079%, indicating a depletion efficiency exceeding 99.3%. These results confirm that the kit achieves highly efficient depletion with excellent specificity, without causing non-specific loss of non-target cells.
Figure 11. Efficient depletion of TCR alpha-beta (αβ)+ cells from human PBMC using the SOLIDEX™-ISOEx Indirect Human TCR alpha-beta (αβ)+ Cell Depletion Kit (Column-based). Results show that the proportion of TCR alpha-beta (αβ)+ cells was 54.7% before depletion (Pre-depletion) and decreased to 0.079% after depletion (After Depletion / Flow-through). This demonstrates that up to 99.3% of TCR alpha-beta (αβ)+ cells were captured and retained by the magnetic separation column, confirming the high specificity of bead capture for TCR alpha-beta (αβ)+ cells.

Background
Why Choose SOLIDEX™-ISOEx Indirect Human TCR alpha-beta (abT)+ Cell Depletion Kit?
- Pristine "Untouched" Cells: The target cells in the flow-through are never bound by antibodies or beads, ensuring zero activation and native phenotype.
- High Depletion Efficiency: The column-based high-gradient magnetic field ensures rigorous trapping of TCR αβ cells, typically removing >99% of contaminants.
- Flexible Strategy: The indirect labeling method allows for high-sensitivity recognition of the TCR αβ complex.
- Downstream Compatibility: Perfect for immediate flow cytometry, functional culture, or molecular analysis.
