Protocol

  1. Preparation: Prepare a single-cell suspension from Human PBMCs (fresh or frozen).
  2. Labeling: Add CD4 Microbeads directly to the cell suspension. Incubate to bind CD4+ T Cells.
  3. Separation: Place a separation column in the magnetic field and apply the cell suspension.
  4. Washing: Magnetically labeled CD4+ cells are retained in the column; unlabeled cells (CD8 T cells, B cells, etc.) are washed away.
  5. Elution: Remove the column from the magnet and elute the purified CD4+ T Cells.

To assess the purity of isolated CD4+ T cells, magnetic separation was performed on human peripheral blood mononuclear cells (PBMCs). Both pre- and post-isolation cells were labeled with CD3-APC and CD4-PE antibodies and subjected to flow cytometric analysis. The purity of CD4+ T cells was significantly enhanced, rising from 28.1% before isolation to 97.0% after isolation, indicating highly efficient enrichment of CD4+ T cells.

What are the sample requirements?
The kit is optimized for fresh or cryopreserved Human PBMCs. It is also highly effective with Leukapheresis products (Leukopaks). Ensure cells are well-dispersed and viable (>90%) for best results.
Do the magnetic beads affect cell activation?
