Protocol

  1. Preparation: Prepare a single-cell suspension from Human PBMCs (fresh or frozen) or Leukopaks.

  3. Separation: Place a separation column in the magnetic field and apply the cell suspension.
  4. Washing: Magnetically labeled CD14+ cells are retained in the column; unlabeled cells (T cells, B cells, NK cells) are washed away.

The SOLIDEX™-ISOEx Human CD14 Isolation Nanobeads (Column-based) was used to isolate and purify CD14+ monocytes from cryopreserved PBMCs. The purity of the cell populations before and after isolation was analyzed by flow cytometry. This experiment achieved a significant increase in CD14+ monocyte purity from 15.2% to 93.7% with a recovery rate of 78.66% as verified by flow cytometry.

FAQ

What are the sample requirements?
This kit is optimized for fresh or cryopreserved Human PBMCs. It is also highly effective with Leukapheresis products (Leukopaks). When using frozen PBMCs, ensure cells are treated with DNase to prevent clumping, which is common with thawing monocytes.
Do the magnetic beads affect cell activation or differentiation?
No. The SOLIDEX™ nanobeads are biodegradable and nano-sized. They are engineered to minimize receptor cross-linking. Validated data shows they do not interfere with the differentiation of monocytes into DCs or Macrophages, nor do they induce artificial cytokine secretion.
How does the Column-based separation work?
The column contains a matrix of ferromagnetic spheres. When placed in a magnetic field, these spheres amplify the magnetic gradient, securely trapping the bead-labeled CD14+ cells. This allows for vigorous washing of contaminants before the target cells are released.
Are the separation columns reusable?
