SOLIDEX™-ISOEx Human CD14 Isolation Nanobeads (Column-based) (Cat No.:GM-Tg-hg-T23212-iso-nanoIMB)
Efficiently isolate Human CD14+ Cells (Monocytes) with SOLIDEX™-ISOEx Nanobeads. Utilizing a robust Positive Selection strategy, this kit specifically targets CD14 antigens to maximize recovery from PBMC or Leukopaks. The Column-based format combined with advanced Magnetic Beads allows for rigorous washing, ensuring High Purity and minimal background contamination. The biodegradable nanobeads are gentle on cells, making them ideal for downstream differentiation into Dendritic Cells (Mo-DCs) or Macrophages without artificial activation.

Application
- Differentiation Studies: Generation of Monocyte-derived Dendritic Cells (Mo-DCs) or Macrophages (M1/M2 polarization).
- Osteoclastogenesis: Differentiation of monocytes into osteoclasts for bone biology research.
- Innate Immunity Research: Analysis of cytokine production (e.g., IL-6, TNF-alpha) upon stimulation.
- Virology: Study of viral entry and replication (e.g., HIV, CMV) in monocytes.
Protocol
- Preparation: Prepare a single-cell suspension from Human PBMCs (fresh or frozen) or Leukopaks.
- Labeling: Add CD14 Microbeads directly to the cell suspension. Incubate to bind CD14+ Monocytes.
- Separation: Place a separation column in the magnetic field and apply the cell suspension.
- Washing: Magnetically labeled CD14+ cells are retained in the column; unlabeled cells (T cells, B cells, NK cells) are washed away.
- Elution: Remove the column from the magnet and elute the purified CD14+ Cells.
Validation Data
High-purity isolation of classical monocytes from human peripheral blood mononuclear cells (PBMC) using the SOLIDEX™-ISOEx Human CD14 Isolation Nanobeads (Column-based)
Catalog No. GM-Tg-hg-T23212-iso-nanoIMB
The SOLIDEX™-ISOEx Human CD14 Isolation Nanobeads (Column-based) was used to isolate and purify CD14+ monocytes from cryopreserved PBMCs. The purity of the cell populations before and after isolation was analyzed by flow cytometry. This experiment achieved a significant increase in CD14+ monocyte purity from 15.2% to 93.7% with a recovery rate of 78.66% as verified by flow cytometry.

Background
Why Choose SOLIDEX™-ISOEx Human CD14 Isolation Nanobeads?
- High Recovery: Positive selection efficiently captures monocytes, ensuring high yields even from donors with lower cell counts.
- Superior Purity: The column matrix acts as a filter, allowing thorough washing of non-specific cells, resulting in higher purity compared to simple tube-based magnetic capture.
- Differentiation Ready: The biodegradable nanobeads do not interfere with downstream differentiation into Dendritic Cells or Macrophages.
- Minimal Activation: Optimized bead size and chemistry prevent artificial activation of monocytes during the isolation process.
