Application
- Cytotoxicity Assays: Isolation of effector cells for tumor cell killing or viral clearance studies.
- CAR-T Cell Development: Source material for engineering Chimeric Antigen Receptor T cells.
- Single-Cell Sequencing: High-recovery isolation for TCR repertoire analysis (scRNA-seq).
- Intracellular Cytokine Staining: Analysis of IFN-gamma, TNF-alpha, or Granzyme B production.
Protocol
- Preparation: Prepare a single-cell suspension from Human PBMCs (fresh or frozen).
- Labeling: Add CD8 Microbeads directly to the cell suspension. Incubate to bind CD8+ T Cells.
- Separation: Place a separation column in the magnetic field and apply the cell suspension.
- Washing: The magnetically labeled CD8+ cells are retained in the column; unlabeled cells (CD4+ T cells, B cells, Monocytes) are washed away.
- Elution: Remove the column from the magnet and elute the highly purified CD8+ T Cells.

Efficient Isolation of CD8+ T Cells Using SOLIDEX™-ISOEx -ISOEx Human CD8 Isolation Nanobeads (Column-Based)
Catalog No. GM-Tg-hg-TA040-iso-nanoIMB
To determine the purity of isolated CD8+ T cell, CD8+ T cells were magnetically sorted from human peripheral blood mononuclear cells (PBMCs). Cells before and after separation were stained with CD3-APC and CD8-PE antibodies and subsequently analyzed by flow cytometry. Results demonstrated a remarkable increase in CD8+ T cell purity from 16.6% pre-isolated to 94.6% post-isolated, confirming highly effective CD8+ T cell enrichment.
Figure 10. Efficient Isolation of CD8+ Cells Using SOLIDEX™-ISOEx -ISOEx Human CD8 Isolation Nanobeads (Column-Based). The T cells were labeled with magnetic nanobeads conjugated with anti-CD8 antibody. And then these cells were enriched by using cell isolation column. The T cells in pre-isolated human PBMCs . T cells with high purity were enriched.

Background
Why Choose SOLIDEX™-ISOEx Human CD8 Isolation Nanobeads?
- Exceptional Recovery: Positive selection captures the maximum number of target CD8+ cells, essential when working with limited patient samples.
- Superior Specificity: The column matrix allows for thorough washing, significantly reducing contamination compared to column-free "clump" methods.
- Biodegradable Nanobeads: The tiny, non-toxic beads degrade naturally and do not over-stimulate the cells, preserving their cytotoxic potential.
- Rapid Workflow: Isolate cells in approximately 45 minutes with a simple, standardized protocol.
