Tissue inhibitor of metalloproteinase-1 (TIMP-1) is a member of the TIMP family, which comprises four structurally related proteins that are known to regulate the activity of matrix metalloproteinases (MMPs). MMPs are zinc-dependent endopeptidases that are involved in the degradation of extracellular matrix (ECM) components, such as collagen, elastin, and proteoglycans. The balance between MMP activity and TIMPs is critical for ECM remodeling, tissue repair, and angiogenesis. TIMP-1 is a secreted glycoprotein that is synthesized by various cells and tissues in animals, including canines/dogs. TIMP-1 has been shown to bind to the active site of MMPs, inhibiting their activity and preventing excessive ECM degradation. In addition to MMP inhibition, TIMP-1 has been shown to have MMP-independent functions, including promoting cell proliferation, differentiation, and apoptosis, as well as modulating immune responses, angiogenesis, and tissue repair processes. TIMP-1 expression is regulated by various cellular processes, including growth factors, cytokines, and ECM components. Its expression has been detected in various cells and tissues in animals, including fibroblasts, endothelial cells, epithelial cells, and macrophages.
