As an enzyme localized primarily within the thyroid gland, TPO takes center stage in the intricate process of thyroid hormone synthesis. TPO's principal role hinges on its enzymatic activity, which catalyzes the iodination of tyrosine residues on thyroglobulin. Thyroglobulin is a substantial glycoprotein synthesized by thyroid follicular cells, serving as the reservoir for thyroid hormones. The iodination process facilitated by TPO is a fundamental step in thyroid hormone formation, converting thyroglobulin into the bioactive hormones thyroxine (T4) and triiodothyronine (T3). These hormones wield commanding influence over metabolic processes, energy expenditure, growth, and various physiological systems in dogs. The mechanism by which TPO operates is intricately tied to iodine utilization. Iodine, an essential trace element acquired through dietary sources, is actively transported into thyroid follicular cells, where it becomes the substrate for TPO. TPO channels iodine atoms onto specific tyrosine residues on thyroglobulin, creating mono- and diiodotyrosines.
