Porcine reproductive and respiratory syndrome virus (PRRSV), also known as blue-ear pig disease virus or porcine arterivirus, is a member of the family Arteriviridae, genus Betaarterivirus. It infects domestic and wild pigs and is the causative agent for two types of diseases: reproductive failure in sows and respiratory disease in piglets and growing pigs.
PRRSV is an enveloped single-stranded positive-sense RNA virus with a non-segmented genome that comprises approximately 15 kilobases. The genome encodes at least 14 proteins, including structural proteins and non-structural proteins. The most significant protein is the viral envelope glycoprotein GP5, which binds to the receptor on host cells and mediates viral entry.
PRRSV infection in pigs primarily occurs via aerosol transmission and direct contact with infected animals, and the virus can persist for prolonged periods in pigs. The incubation period of PRRSV ranges from two to six days, and the duration of the disease can vary from acute to chronic.
Reproductive failure due to PRRSV can result in abortions, stillbirths, mummification of fetuses, and weak or malformed piglets. Sows infected with PRRSV have a higher risk of developing secondary bacterial infections such as postpartum dysgalactia syndrome, which leads to a reduction in milk production. In contrast, respiratory disease caused by PRRSV leads to fever, coughing, and difficulty breathing in affected animals. The severity of the respiratory disease can range from mild to severe, with morbidity rates reaching 100% and mortality rates ranging from 5% to 50%.
To detect PRRSV infection in pigs, several diagnostic methods are available. Serological tests, such as enzyme-linked immunosorbent assay (ELISA) and immunofluorescence assay (IFA), are commonly used to detect antibodies against PRRSV. These tests are useful for detecting past exposure to the virus but cannot detect acute infections.
Nucleic-acid based tests, including reverse transcription polymerase chain reaction (RT-PCR) and loop-mediated isothermal amplification (LAMP), can detect viral RNA in infected animals. RT-PCR is a highly sensitive and specific method that can detect low levels of viral RNA. The LAMP technique is rapid, sensitive, and does not require sophisticated equipment, making it ideal for field use.
Virus isolation is another method used to detect PRRSV infection. This method involves culturing the virus in cell culture systems, followed by identification and characterization of the virus using serological and molecular techniques.
