    S100-A9, also known as calgranulin B, is a calcium-binding protein belonging to the S100 protein family, renowned for its involvement in a myriad of cellular processes, particularly those related to inflammation and immune responses. In dogs (canines), S100-A9 is a key player in innate immunity, primarily produced by neutrophils and monocytes. The significance of S100-A9 lies in its ability to form a heterodimeric complex with S100-A8, collectively known as S100A8/A9 or calprotectin. This complex serves as a critical component of the innate immune system's arsenal, playing multifaceted roles in immune regulation, host defense, and tissue homeostasis. The release of S100A8/A9 occurs primarily in response to inflammatory cues. Activated neutrophils and monocytes release this complex into the extracellular milieu as part of the innate immune response. S100A8/A9, once externalized, acts as a potent pro-inflammatory mediator, capable of initiating and amplifying immune responses. Its functions include the recruitment of immune cells to sites of infection or injury and the facilitation of phagocytosis, a crucial process by which immune cells engulf and eliminate pathogens. Additionally, S100-A9 has been implicated in other cellular processes beyond inflammation. It plays a role in cell adhesion, differentiation, and tissue repair. As a damage-associated molecular pattern (DAMP) molecule, S100-A9 serves as a sentinel, alerting the immune system to tissue damage or infection.
