    Bovine Herpes Virus 1 is an alphaherpesvirus that predominantly infects cattle, and occasionally sheep and goats. BHV-1 is particularly prevalent in countries with intensive beef and dairy cattle industries, and can have significant economic impact due to reduced milk production and loss of meat quality.

    BHV-1 is a double-stranded DNA virus composed of four main structural components- the capsid, tegument, envelope, and viral genome. The capsid is composed of multiple copies of the major capsid protein, which encloses the viral genome and protects it from environmental damage. The tegument, located between the capsid and envelope, contains various viral proteins that help facilitate viral entry into host cells and evade the host immune response. The viral envelope is derived from the host cell membrane during replication and contains a mixture of viral and host proteins, including glycoprotein D (gD).

    The most important gene of BHV-1 is gD, which encodes one of the envelope glycoproteins responsible for facilitating viral entry into host cells. The gD receptor complex consists of several host proteins, including nectin-1 and herpesvirus entry mediator (HVEM), which initiate the fusion process between the viral and host membranes. Other important genes include thymidine kinase (TK) and immediate early (IE) genes. TK is involved in viral DNA synthesis and is targeted by antiviral drugs, while IE genes play a role in regulating viral gene expression.

    BHV-1 primarily infects cattle via respiratory routes, causing clinical symptoms such as fever, nasal discharge, cough, and conjunctivitis. Infection can also occur via genital routes, leading to vulvovaginitis and balanoposthitis. Although sheep and goats are susceptible to BHV-1 infection, the clinical manifestations are generally milder than in cattle.

    Diagnosing BHV-1 infection requires a combination of clinical observation and laboratory tests. Diagnostic methods include virus isolation, PCR, and ELISA, which can detect viral DNA or antigens in various tissue samples such as nasal swabs, ocular secretions, or genital fluids. PCR and ELISA assays target specific genes or proteins, most commonly gD or TK. Serological tests, such as the serum neutralization assay, detect antibodies against BHV-1 in blood samples, indicating previous exposure or vaccination.

    Prevention and control of BHV-1 infection is essential to minimize its economic consequences. Vaccination with modified-live or killed vaccines is the most effective strategy for controlling BHV-1 in cattle herds. Modified-live vaccines provide longer-lasting immunity and are preferred for use in breeding females to prevent vertical transmission to their offspring. In addition to vaccination, reducing stress and implementing proper biosecurity measures can help lower the incidence of BHV-1 infection within cattle herds.
