    Ornithobacterium rhinotracheale (ORT) is a Gram-negative bacterium that belongs to the family Pasteurellaceae. It is a respiratory pathogen of poultry species, causing serious conditions such as avian infectious bronchitis (IB), avian bordetellosis, and avian coryza. Infections with ORT can lead to several clinical signs which include nasal discharge, respiratory distress, coughing, sneezing, and decreased egg production in infected birds.

    ORT was first isolated in the 1980s from turkeys with respiratory disease, and since then, it has been reported in various parts of the world, including Europe, Asia, South Africa, and America. This wide distribution of ORT may be attributed to its ability to survive in different environments and hosts. It's worth noting that this pathogen is specific to birds and does not pose any significant threat to humans.

    ORT is a small, non-motile bacterium that measures approximately 1-2 μm in length and 0.5 μm in diameter. Its cell wall contains peptidoglycan, which provides structural support, and lipopolysaccharides (LPS), which are essential for virulence. ORT also has an outer membrane that is made up of various proteins, with the most prominent being the outer membrane protein A (OmpA). The OmpA protein is involved in the pathogenicity of ORT by promoting bacterial adhesion to host cells.

    One of the diagnostic methods used to identify ORT infections is through culture. ORT grows best on blood agar plates at a temperature range of 37-42°C, where colonies appear after 24-48 hours of incubation. However, culturing ORT may be difficult due to its fastidious growth requirements, and as a result, other diagnostic methods have been developed to identify the pathogen.

    Polymerase Chain Reaction (PCR) is a molecular diagnostic tool that has been used to successfully detect ORT infections. PCR amplifies specific genes found within the bacterial genome. For example, the 16S rRNA gene has been used as a target in identifying ORT strains. Besides the 16S rRNA gene, other genes such as the sodA gene and hsf have also been used as targets for diagnosing ORT infections.

    Enzyme-linked immunosorbent assay (ELISA) is another commonly used diagnostic tool for the detection of ORT infections. ELISA can detect specific antigens or antibodies present within infected tissues, fluids, or sera. In ORT, the OmpA protein and LPS are the primary targets used in ELISA-based diagnosis. A positive diagnosis in ELISA indicates the presence of these proteins within the sample being analyzed.

    Immunohistochemistry (IHC) is another diagnostic method used for detecting ORT infections. IHC is a technique that uses antibodies to detect antigens within infected tissues. Like ELISA, IHC primarily targets antigens associated with ORT, such as OmpA and LPS.
