TCRαβ+ Cells refer primarily to T cells that express αβ T cell receptors (TCR), including CD4+ helper T cells and CD8+ cytotoxic T cells. These cells make up more than 95% of the peripheral blood T cells and are a core component of the adaptive immune system.
TCRαβ is a heterodimer receptor on the surface of T cells, composed of α and β chains, specifically expressed on conventional T cells but not on γδ T cells or non-T cells, providing highly specific antigen recognition. Clinical Applications:
In hematopoietic stem cell transplantation (HSCT), studies show that removing αβ T lymphocytes from peripheral blood stem cells (PBSC), while retaining all other cell populations, supports allogeneic transplantation by providing immune modulation and reducing transplant rejection responses. Additionally, TCRαβ/CD19 depletion in hematopoietic stem cell transplantation has shown promising applications in treating hematologic malignancies. Finally, a clinical trial studied the effect of TCRαβ+ and CD19+ cell depletion on enhancing γδ T cell cytotoxicity against leukemia cells in the presence of ZOLE (Zoledronic acid), with results showing that T cell reconstitution played a positive regulatory role during treatment.

Efficient depletion of TCR alpha-beta (αβ)+ T cells from human PBMC using the SOLIDEX®-ISOEx Indirect Human TCR alpha-beta (αβ)+ Cell Depletion Kit (Column-based)

This experiment was designed to evaluate the depletion efficiency of the SOLIDEX®-ISOEx Indirect Human TCR alpha-beta (αβ)+ Cell Depletion Kit (Column-based) for removing TCR alpha-beta (αβ)+ cells. Data analysis demonstrated the kit's excellent depletion performance. In the initial PBMC sample, TCR alpha-beta (αβ)+ cells accounted for 54.7%. After processing through the separation column, the residual TCR alpha-beta (αβ)+ cells in the flow-through fraction dramatically decreased to 0.079%, indicating a depletion efficiency exceeding 99.3%. These results confirm that the kit achieves highly efficient depletion with excellent specificity, without causing non-specific loss of non-target cells.
