Introduction of SOLIDEX™-ISOEx CD14+ Cell Isolation Solution
CD14+ cells primarily refer to monocytes, which are key immune cells in peripheral blood and bone marrow, comprising about 5-10% of the white blood cells. Monocytes play an essential role in innate immunity, differentiating into macrophages or dendritic cells, participating in pathogen phagocytosis, antigen presentation, cytokine secretion (such as IL-6 and TNF-α), and inflammation regulation.
The CD14 antigen is part of the LPS receptor complex. Antibody binding to CD14 does not trigger signal transduction, as CD14 lacks a cytoplasmic domain. CD14 is strongly expressed on most monocytes and macrophages, with weaker expression on neutrophils and some myeloid dendritic cells. Research Applications:
Used to generate macrophages ex vivo, or for cytotoxicity studies and migration assays to assess their function in inflammatory models, such as phagocytic activity or chemotactic factor-induced migration mechanisms.
Clinical Applications:
Positive selection of CD14+ monocytes is used for differentiating into dendritic cells (DCs) for maintenance therapy in pleural mesothelioma patients. Additionally, the method is used to prepare clinical-grade DC vaccines, where autologous DCs loaded with CMV pp65 mRNA have been shown to be safe and feasible in healthy volunteers and HSCT recipients, inducing CMV-specific T cell responses, thereby supporting viral infection prevention.

Used to study the key role of monocytes in regulating immune molecule expression in pathogenic responses.
Clinical Applications:
Depletion of CD14+ cells followed by positive selection of CD4+ T helper cells from PBMCs is used for efficient separation of CD4+ T helper cells with strong expression, supporting T cell function analysis or transplantation optimization. Moreover, CD14/CD25 depletion removes myeloid cells and Tregs, enhancing CAR-T cell efficacy in the treatment of refractory/relapsed non-Hodgkin lymphoma and reducing immune suppression. In cGMP large-scale production, depletion of CD14+ cells ensures successful CAR-T cell transduction and expansion, increasing production purity and activity.

High-purity isolation of classical monocytes from human peripheral blood mononuclear cells (PBMC) using the SOLIDEX™-ISOEx Human CD14 Isolation Nanobeads (Column-based)

The SOLIDEX™-ISOEx Human CD14 Isolation Nanobeads (Column-based) was used to isolate and purify CD14+ monocytes from cryopreserved PBMCs. The purity of the cell populations before and after isolation was analyzed by flow cytometry. This experiment achieved a significant increase in CD14+ monocyte purity from 15.2% to 93.7% with a recovery rate of 78.66% as verified by flow cytometry.

High-purity isolation of classical monocytes from human peripheral blood mononuclear cells (PBMC) using the SOLIDEX™-ISOEx Untouched Human Classical Monocyte Isolation Kit (Column-based)

In this experiment, the SOLIDEX™-ISOEx Untouched Human Classical Monocyte Isolation Kit (Column-based) was used to isolate and purify classical monocytes from cryopreserved PBMCs. First, non-monocytic cells were specifically labeled with a biotin-conjugated antibody cocktail (Antibody Cocktail), followed by incubation with anti-biotin nano-magnetic beads. The purity of the cell populations before and after isolation was analyzed by flow cytometry. The purity of CD14+ classical monocytes in the flow-through fraction increased from 21.7% to 92.1%. The experimental results demonstrate that this kit effectively removes contaminating cells, yielding high-purity, untouched (antibody-unbound) classical monocytes.
