SOLIDEX™ NK Cell Isolation Solution
Introduction of SOLIDEX™ NK Cell Isolation Solution
Natural Killer (NK) Cells are core effector cells of the innate immune system, capable of killing virus-infected cells or cancer cells without prior sensitization. They play a crucial role in antiviral defense, antitumor immunity, and transplant immunity. Key surface markers of NK cells include CD56 (Neural Cell Adhesion Molecule, NCAM) and CD16 (FcγRIII, low-affinity Fc receptor).
CD56 promotes the adhesion and recognition of target cells, enhancing cytotoxic activity. Magnetic bead sorting is a commonly used technique for isolating NK cells, utilizing beads coated with antibodies to achieve efficient separation. GeneMedi provides three types of magnetic bead selection: Negative Selection/Untouched, Positive Selection, and Depletion, to meet various research and therapeutic needs.
Isolation method of SOLIDEX™ NK Cell Isolation Solution
- Positive Selection
- Negative Selection
- Depletion
Research Applications:
Positive selection is used to enrich CD56+ NK cells and is widely applied in research on the development, proliferation, cytotoxic activity, and immune functions of NK cells. Using CD56 magnetic beads, researchers can deeply explore the key roles of NK cells in immune surveillance, particularly in cancer immunotherapy.
Clinical Applications:
In immunotherapy, positive selection is used for the enrichment of clinical-grade NK cells. After removing CD3+ T cells, CD56 magnetic beads are employed to isolate high-purity NK cells. Additionally, positive selection is also used in the preparation of CAR NK cell strategies. Studies have shown that CAR-NK cells (expressing anti-CD123 CAR) specifically kill CD123-positive tumor cells.

Research Applications:
Negative selection enriches pure NK cells by removing non-target cells. This technique is used to study NK cell proliferation, cytotoxicity, and their role in tumor immunity. It is also used to analyze the function of NK cell surface receptors and cytokine secretion, helping to elucidate the mechanisms of NK cells in immune responses.

Research Applications:
The depletion method involves the removal of NK cells to study their role in immune responses, especially in the context of tumor immune evasion mechanisms. This method aids in exploring the critical functions and regulation of NK cells in immune responses.
Clinical Applications:
Depletion technology is used in clinical research to assess the impact of NK cells on disease progression, particularly in hematological diseases and hematopoietic stem cell transplantation. By depleting NK cells, the function of stem cells can be improved, enhancing the success of transplant treatments.
