Introduction of SOLIDEX™-ISOEx Pan-T Cell Isolation Solution

Pan-T Cells encompass all T cell subtypes, including helper T cells, cytotoxic T cells, and regulatory T cells. They are core to adaptive immunity, coordinating cell-mediated immunity and humoral immune regulation, playing a crucial role in infection defense, tumor immunity, and autoimmune diseases.

CD3E (CD3 epsilon) is a key subunit of the T cell receptor (TCR) complex, part of the CD3 molecule family (which includes CD3ε, CD3δ, CD3γ, and CD3ζ subunits), and is widely expressed on the surface of all mature T cells, making it a universal marker for Pan-T cells. immunomagnetic cell separation is a commonly used technique for isolating Pan-T cells, utilizing beads coated with antibodies for efficient separation. Research Applications:

The depletion method, by depleting T cells, allows for the study of the function of other immune cells, especially innate immune cells (such as NK cells) and their anti-infective roles without T cell interference. Additionally, by removing T cells, researchers can focus on studying the immune functions of non-T cell populations and their roles in disease.

Clinical Applications:

Studies show that using CD3/CD19 depletion grafts with RIC treatment leads to delayed reconstitution of T cells and B cells, while NK cell reconstitution occurs earlier and more rapidly. Moreover, when comparing CD34+ selection grafts with CD3/CD19 depletion grafts for immune reconstitution in pediatric acute leukemia patients undergoing haploidentical stem cell transplantation, CD3/CD19 depletion grafts were found to better promote immune reconstitution. In clinical applications involving NK cells, selecting CD56+ cells after depleting CD3+ cells effectively enriches clinical-grade NK cells, providing support for implementing CAR-NK cell strategies.

Efficient Isolation of Pan-T Cells Using SOLIDEX™-ISOEx Human CD3 Isolation Nanobeads (Column-Based)

To assess the purity of isolated T cells, CD3+ T cells were magnetically sorted from human peripheral blood mononuclear cells (PBMCs). Cells before and after separation were stained with APC-conjugated CD3 antibody and propidium iodide (PI), followed by flow cytometric analysis. The results demonstrated a significant increase in CD3+ T cell purity from 52.6% pre-isolated to 95.0% post-isolated, confirming successful T cell isolation.

Efficient Isolation of Pan-T Cells Using SOLIDEX™-ISOEx Human CD3 Isolation Nanobeads (Column-Free)

To evaluate the purity of isolated T cells, CD3+ T cells were magnetically sorted from human peripheral blood mononuclear cells (PBMCs). Cells before and after separation were stained with APC-conjugated CD3 antibody and analyzed by flow cytometry. The CD3+ T cell purity demonstrated a substantial increase from 65.9% pre-separation to 98.6% post-separation, confirming highly effective T cell isolation.

High-purity isolation of Pan-T cells from human PBMC using the SOLIDEX™-ISOEx Untouched Human Pan-T Cell Isolation Kit (Column-based)

In this experiment, the SOLIDEX™-ISOEx Untouched Human Pan-T Cell Isolation Kit (Column-based) was used to isolate and purify Pan-T cells from human peripheral blood mononuclear cells (PBMC). First, non-Pan-T cells were specifically labeled with a biotin-conjugated antibody cocktail (Antibody Cocktail), followed by incubation with anti-biotin nano-magnetic beads. When the cell suspension passed through the separation column, the magnetically labeled non-target cells were retained in the column, while the unlabeled, untouched Pan-T cells flowed out with the elution buffer. Data showed that the proportion of Pan-T cells increased significantly from 58.9% before isolation to 96.1% after isolation. The experimental results demonstrate that this kit effectively removes various types of non-Pan-T cells, yielding high-purity Pan-T cells that retain their original biological activity.
