    Dirofilaria immitis is a parasitic nematode that infects primarily dogs, but also cats and wild canids such as foxes, wolves, and coyotes. It is transmitted by mosquitoes that carry infective third-stage larvae, which enter the host's bloodstream when the insect bites. Once inside the host, the larvae migrate to the right side of the heart and pulmonary arteries, where they develop into adult worms and mate, producing microfilariae that circulate in the bloodstream and can be picked up by mosquitoes.

    Dirofilaria immitis has a unique morphology characterized by a long and slender body with a pointed tail. The adult worms can reach up to 30 cm in length and are often coiled in the pulmonary arteries, causing obstruction and damage to the heart and lungs. The pathogen is classified as eukaryotic and belongs to the phylum Nematoda.

    The genome of Dirofilaria immitis contains several genes responsible for its survival within the host. The pathogen produces an enzyme called urease, which is involved in metabolism, and helps the pathogen adjust to the host's acidic environment. In addition, the pathogen produces several proteins such as heat shock proteins and antioxidant enzymes, which aid in survival and persistence within the host's body.

    Heartworm disease caused by Dirofilaria immitis can have significant health consequences if left untreated. Infected animals may experience symptoms such as coughing, lethargy, and weight loss. Heart failure may occur in severe cases, leading to pulmonary hypertension and eventually death. The severity of the disease depends on several factors, such as the number of worms present, the duration of infection, and the animal's immune response.

    Various diagnostic methods are available for detecting Dirofilaria immitis infection. The most common diagnostic tests include antibody tests, antigen tests, and nucleic acid tests. These methods target specific genes or proteins, such as the 16S ribosomal RNA gene, the Wolbachia surface protein, or the dihydrofolate reductase-thymidylate synthase (DHFR-TS) gene. Examples of diagnostic tests include the Knott test, the modified Knott test, and the polymerase chain reaction (PCR) test.

    The Knott test is a basic microscopic test that involves concentrating microfilariae from a blood sample using a staining solution. The modified Knott test is a slight variation of the Knott test that uses a different staining solution and adds an additional centrifugation step to increase the sensitivity of the test. Both tests are relatively easy to perform and inexpensive but may not be as sensitive as other methods.

    Antigen tests, on the other hand, detect the presence of adult female worm antigens in the bloodstream. These tests are more accurate than microfilarial tests but may have limitations in detecting very light infections or male-only infections.

    Nucleic acid tests, such as the PCR test, detect the genetic material of the pathogen, making them the most sensitive and specific diagnostic tests available. These tests require specialized equipment and expertise, but they can detect the infection early and distinguish between active, past, and false-positive infections.
