CD45RA+ cells primarily refer to naive T cells and certain other leukocytes (e.g., subsets of B cells and NK cells), widely present in peripheral blood, bone marrow, and lymphoid tissues. CD45RA, an isoform of the CD45 protein, is specifically expressed on the surface of naive T cells and other non-activated leukocytes, distinguishing them from memory T cells (which express CD45RO) with high specificity. Research Applications:

Depletion of CD45RA+ cells is commonly used to isolate CD45RO+ cells from pre-sorted CD4 or CD8 T cells, facilitating studies of memory T cell functions and immunological characteristics to further understand their role in immune responses.

Clinical Applications:

Depletion of CD45RA+ cells from peripheral blood stem cell (PBSC) products significantly enriches CD4+ and CD8+ central memory T cells, which can proliferate and produce effector cytokines in response to opportunistic pathogens, enhancing immune reconstitution and effectively reducing CD8-mediated allogeneic reactivity. Studies also demonstrate that CD45RA-depleted donor lymphocyte infusion combined with post-transplant cyclophosphamide effectively improves clinical outcomes.
