CD34+ cells primarily refer to hematopoietic stem cells (HSCs) and hematopoietic progenitor cells (HPCs), which are found in the bone marrow, umbilical cord blood, and peripheral blood. Research Applications:
Positive selection of CD34+ cells for in vitro culture is used to analyze the effects of various tyrosine kinase inhibitors on downstream targets. Subsequent techniques such as Western blot, qPCR, and cell cycle analysis are employed to investigate the molecular mechanisms of these cells. Additionally, positive selection is frequently used in studies related to hematopoiesis, differentiation, colony formation, and the expression of surface markers.
Clinical Applications:
Stem cell grafts obtained through positive selection are highly purified, free from other cell populations, ensuring the safety of transplantation. In the treatment of acute myeloid leukemia (AML), transplantation of these purified CD34+ stem cells can effectively alleviate the disease. Moreover, this technique is also applied in the treatment of patients with non-union fractures, showing promising clinical prospects.
