Canine p-tau212 and p-tau214, the phosphorylated Tau Proteins unique to dogs, occupy a pivotal role within the intricate landscape of canine neurobiology. At their core, they are Tau proteins that have undergone specific biochemical modifications – phosphorylation – at residues 212 and 214. Normally, Tau proteins in dogs play a vital role in stabilizing microtubules, essential for axonal stability and intracellular transport within neuronal cells.
However, in the context of neurodegenerative diseases, the narrative takes a different course. Phosphorylation events can trigger the misfolding and aggregation of Tau proteins, and in dogs, p-tau212 and p-tau214 tend to aggregate and form neurofibrillary tangles (NFTs) within neural tissues.
These NFTs, akin to those observed in human neurodegenerative diseases such as Alzheimer's, represent a pathological signature of various canine neurodegenerative conditions. The accumulation of p-tau212 and p-tau214 in NFTs contributes to progressive neuronal dysfunction, cognitive decline, and motor disturbances characteristic of conditions such as canine cognitive dysfunction syndrome (CCDS) and specific encephalopathies.
