Q1: Can the SOLIDEX®-ISOEx Magnetic Sorter series be used for either positive or negative selection?

Yes, when used with our column-free nanobeads, this series fully supports both Positive Selection, Negative Selection and Depletion.

Q2: How does the isolation work?

Mechanism: After target or non-target cells are labeled with specific antibodies and magnetic particles, the sample tube is placed into the SOLIDEX®-ISOEx Magnetic Sorter. The strong magnetic field rapidly pulls the magnetically labeled cells to the wall of the tube.

  • For positive selection, the targeted cells on the wall are retained while the supernatant is poured off.

  • For negative selection, the supernatant containing the untouched target cells is collected.

Q3: Which columns do I use?

This series is entirely Column-Free. You do not need to use or purchase any column consumables.

Q4: How do I choose between Sorter-5, Sorter-15, and Sorter-50?

The choice of sorter depends strictly on your sample volume and the type of tube used:

  • SOLIDEX®-ISOEx Magnetic Sorter-5: Designed for standard 5 mL flow cytometry tubes (e.g., 12 × 75 mm tubes).

  • SOLIDEX®-ISOEx Magnetic Sorter-15: Accommodates 14 mL or 15 mL standard centrifuge tubes, ideal for medium-volume samples.

  • SOLIDEX®-ISOEx Magnetic Sorter-50: Fits 50 mL conical centrifuge tubes, specifically designed for large-volume samples or high cell concentrations.

Q5: How does the binding of the magnetic particles affect the cells?

The biological function of the cells remains completely unaltered. Our magnetic particles are biologically inert and do not induce abnormal activation or phenotypic changes upon binding.

Q6: Are the particles FACS-compatible?

Because the SOLIDEX®-ISOEx Column-Free system utilizes larger 100 nm nanobeads to ensure robust magnetic capture (unlike the 50 nm beads used in column-based systems), the increased particle size can interfere with the light scatter signals during flow cytometry following positive selection. Therefore, to ensure accurate FACS analysis and reliable sensitive downstream assays, we highly recommend the following:

  1. Prioritize Negative Selection: This strategy allows you to obtain "untouched," completely bead-free target cells that can be directly analyzed via FACS.

  2. Bead Removal for Positive Selection: If positive selection is required, please ensure the magnetic particles are removed from the cell surface using our dedicated release protocol prior to FACS analysis or functional assays.

Q7: Can I alter the isolation time in the magnet?

We do not recommend altering the protocol. It is strongly advised to adhere strictly to the isolation times specified in the protocol. Shortening the time leads to incomplete magnetic capture (reducing yield), while over-extending it may increase non-specific binding.

Q8: For positive selection, can I perform more than 3 isolations to increase purity?

For positive selection, 1 to 3 washes typically achieve optimal purity. While performing more than 3 washes might incrementally increase purity, it will inevitably cause mechanical stress to the cells, leading to decreased cell viability and a significant drop in overall recovery yield.

Q9: How can I analyze the purity of my enriched sample?

Purity is best analyzed via Flow Cytometry. We recommend staining the enriched cells with a fluorochrome-conjugated antibody that targets a different epitope than the one used for isolation to prevent false-negative results caused by steric hindrance.
