B Cells (B lymphocytes) are central to humoral immunity, responsible for producing antibodies to fight pathogens and assisting T cell activation through antigen presentation. They play a crucial role in infection defense, vaccine responses, and autoimmune diseases. Key surface markers of B cells include CD19 and CD20.
CD19 acts as a co-receptor for the B Cell Receptor (BCR), enhancing BCR signaling and promoting B cell activation, proliferation, and differentiation into plasma cells to produce antibodies. Defects in CD19 can lead to immunodeficiency disorders. immunomagnetic cell separation is a commonly used technique for isolating B cells, utilizing beads coated with antibodies for efficient separation. Research Applications:
Negative selection isolates untouched B cells, suitable for studies on BCR signaling, antibody secretion, vaccine responses, in vitro plasma cell differentiation, or CRISPR gene editing (e.g., BCR knockout). This method is ideal for experiments involving antibody affinity maturation. It can also be used to further isolate B cell subsets, such as analyzing transgene expression in transgenic models via Northern blot analysis.

Research Applications:
Depletion is used to enrich non-B cells (e.g., T cells, NK cells, with purity of 85-95%) for studies on non-B cell functions or disease models where B cells need to be reduced, such as in autoimmune diseases.
Clinical Applications:
Clinically, CD19 is often depleted in combination with CD3 or TCRαβ. For example, CD3/CD19 depletion grafts have been shown to delay T/B cell reconstitution after HHCT while accelerating NK cell recovery, enhancing antitumor activity. TCRαβ/CD19 depletion in URD HSCT is used for pediatric hematological malignancies, reducing the risk of GVHD and EBV infection.
