CD8+ T cells (cytotoxic T cells) are key effector cells of the adaptive immune system, playing a central role in antiviral and antitumor immunity by directly lysing virus-infected or cancerous cells. Their primary surface marker, CD8, is co-expressed with CD3.
CD8 acts as a co-receptor, binding to MHC class I molecules to enhance T cell receptor (TCR) recognition of antigens on infected or tumor cells, promoting the release of granzymes and perforins to induce target cell apoptosis, and secreting cytokines such as IFN-γ to amplify inflammatory responses. Research Applications:
Positive selection enriches CD8+ T cells for functional studies, including proliferation assays, cytotoxicity assays, and analysis of in vitro cytokine production (e.g., IFN-γ, TNF-α). It also facilitates research on viral infections, particularly the role of CD8+ T cells in antiviral immune responses, enabling detailed investigation of their effector mechanisms.
Clinical Applications:
Positive selection is critical for CAR-T cell therapy, enabling the isolation of CD4+ and CD8+ T cells to produce GD2-CAR-T cells, which demonstrated significant clinical and radiological improvements in three out of four patients in a phase I trial. It also supports preclinical development of antigen-induced IL-18-enhanced CAR-T cells for GD2+ solid tumors and enables large-scale automated T cell production, facilitating industrialization and standardization of T cell therapies.

Negative selection purifies CD8+ T cells by removing non-CD8+ cell populations, allowing isolation of specific cytotoxic T cell subsets, such as naive, memory, or activated CD8+ T cells. Research Applications:
Depletion of CD8+ T cells from peripheral blood mononuclear cells (PBMCs) is used in severe combined immunodeficiency (SCID) mouse models to study immune reconstitution and the functional recovery of non-CD8+ immune components.
Clinical Applications:
