Introduction of SOLIDEX®-ISOEx CD8-T Cell Isolation Solution

CD8+ T cells (cytotoxic T cells) are key effector cells of the adaptive immune system, playing a central role in antiviral and antitumor immunity by directly lysing virus-infected or cancerous cells. Their primary surface marker, CD8, is co-expressed with CD3.

CD8 acts as a co-receptor, binding to MHC class I molecules to enhance T cell receptor (TCR) recognition of antigens on infected or tumor cells, promoting the release of granzymes and perforins to induce target cell apoptosis, and secreting cytokines such as IFN-γ to amplify inflammatory responses. Research Applications:

Positive selection enriches CD8+ T cells for functional studies, including proliferation assays, cytotoxicity assays, and analysis of in vitro cytokine production (e.g., IFN-γ, TNF-α). It also facilitates research on viral infections, particularly the role of CD8+ T cells in antiviral immune responses, enabling detailed investigation of their effector mechanisms.

Clinical Applications:

Positive selection is critical for CAR-T cell therapy, enabling the isolation of CD4+ and CD8+ T cells to produce GD2-CAR-T cells, which demonstrated significant clinical and radiological improvements in three out of four patients in a phase I trial. It also supports preclinical development of antigen-induced IL-18-enhanced CAR-T cells for GD2+ solid tumors and enables large-scale automated T cell production, facilitating industrialization and standardization of T cell therapies.

Research Applications:

Negative selection purifies CD8+ T cells by removing non-CD8+ cell populations, allowing isolation of specific cytotoxic T cell subsets, such as naive, memory, or activated CD8+ T cells. These subsets are used to study antigen-specific responses and immune memory formation in infection or tumor models.

Research Applications:

Depletion of CD8+ T cells from peripheral blood mononuclear cells (PBMCs) is used in severe combined immunodeficiency (SCID) mouse models to study immune reconstitution and the functional recovery of non-CD8+ immune components.

Clinical Applications:

CD8+ T cell depletion from healthy donor leukapheresis, followed by enrichment of CD25high cells, enables isolation of polyclonal regulatory T cells (Tregs) from total nucleated cells. These Tregs are suitable for bedside transfusion and support high-throughput clinical-scale Treg production and safe, automated manufacturing of chimeric antigen receptor Tregs (CAR-Tregs) for immunotherapy applications.

Efficient Isolation of CD8+ T Cells Using SOLIDEX®-ISOEx Human CD8 Isolation Nanobeads (Column-Based)

To determine the purity of isolated CD8+ T cell, CD8+ T cells were magnetically sorted from human peripheral blood mononuclear cells (PBMCs). Cells before and after separation were stained with CD3-APC and CD8-PE antibodies and subsequently analyzed by flow cytometry. Results demonstrated a remarkable increase in CD8+ T cell purity from 27.4% pre-isolated to 96.0% post-isolated, confirming highly effective CD8+ T cell enrichment.

Efficient Enrichment of High-Purity CD8+ T Cells from Human PBMCs Using SOLIDEX®-ISOEx Untouched Human CD8+ T Cell Isolation Kit (Column-based)

To evaluate the isolation efficiency of the SOLIDEX®-ISOEx Untouched Human CD8+ T Cell Isolation Kit (Column-based), CD8+ T cells were enriched from human peripheral blood mononuclear cells (hPBMCs) via negative selection. Non-target cells were labeled with a biotinylated antibody cocktail and depleted using magnetic beads within a separation column.
Cell fractions were stained with CD3-APC and CD8-PE for flow cytometric analysis. The frequency of CD8+ T cells demonstrated a substantial increase from 22.6% in the pre-isolation sample to 91.9% in the post-isolation fraction. These results confirm that the column-based system provides superior depletion of non-target populations, yielding highly purified "untouched" CD8+ T cells.
